In order to enter the locked room in Sheriff’s house, you have to go to the Nonde Saloon that is nearby. Walk into it and then look for a bookcase. When you find it, simply interact with it and the bookcase will open up the way to the room. Now it is important to note that you need to get all the collectibles in the Bounty of Blood DLC.
